Molecular Identification of Coffea Arabica Varieties in Nepal: Insights from Phylogenetic Analysis

This study aimed to identify and characterize the coffee varieties cultivated in Nepal using molecular phylogenetic analysis. The identification genetic relationship of twenty five were collected from Coffee Development Center, Gulmi, Nepal. DNA was isolated leaf tissue, Internal Transcribed Spacers Region (ITS)-specific PCR performed, followed by sequencing tree construction. BLASTN performed similarities with sequences National Center for Biotechnology Information (NCBI) Database. Evolutionary divergence between computed Maximum Composite Likelihood Model. Sequences analyzed Model Tamura-Nei model construct phylogeny. phylogeny confirm all samples be Coffea arabica. pairwise comparison found 0% 4.3%. Divergence 4.3% detected CDC-S21 CDC-S73. With this, we identified C. arabica also relatedness among our varieties.
